I think MAME search for games in a sub-folders with the same name as the software list name, so, you can set only this in your mame.ini:
d:\mame\roms;c:\emulator\mame\softwarelist
and run a Atari 2600 game, asteroids.
mame64.exe a2600 asteroids
MAME will search for asteroids.zip in the above folders but also in (no need to set these in mame.ini):
d:\mame\roms\a2600\
c:\emulator\mame\softwarelist\a2600\
But I don't think MAME supports recursive folders search... any sub-folder inside the folders you set in mame.ini.
For frontend writers that do games auditing inside the frontend (me included), this would be a nightmare to support.
