No version requires a VMU to unlock stuff. As players play the game, they win experience points which are added to a counter in the game (in fact it's stored on the NAOMI motherboard). The game "levels up" and new characters and new stages are unlocked when a certain value is reached. Japanese and American versions work the same way.
Using a VMU, the player can win points on the home version and directly use characters who may not have been unlocked yet on the arcade machine. But it's not mandatory since as times goes on, the machine will eventually gain enough points to have all characters unlocked.
The operator can level up the machine artificially using commands input in test mode. It works with both Japanese and English versions of the game but the codes to enter are different.
