To help, we need answers to these simple questions: Which operating system? WinXP? Win7? OSX? Linux? Which MAME variant? Windows MAME? SDLMAME? MAMEUI? MAMEUIFX? WolfMAME? What kind of controllers? PS3 USB controllers? HotRod? X-Arcade? How do the controllers show up in your OS? Like keyboard devices? High-level keystroke events? HID game controller? If you have controllers that emulate keyboard devices, you can fix your problem by either mapping each one to different keystrokes, or enabling multi-keyboard support.
