I've had this question for a while and normally get around it by deleting "nplayers.ini".
But when I have it in place and launch MAMEUI64, I get the following pop-up box:
Error parsing folders\nplayers.ini: missing [folder name] or [ROOT_FOLDER]
Is there something else that I have to do with it? (I'm placing it in the folders directory).

For MAMEUI64, you have to use Multiplayer.ini (from NPlayers32 archive), not nplayers.ini.
Please tell me if it resolves your issue.

Thanks. I deleted nplayers and put in multiplayer and the error is no more.
