Creating a dos bootable USB stick

I am in need of some help creating a bootable DOS usb stick.
I can create a bootable stick ok (using "rufus") however failing to work when adding mame directories etc
Any help very much appreciated.

Google HP USB disk formatting tool and try using that instead of Rufus.

You'll have to provide a bit more information than just "failing to work".
