What get me is that I can't get the menu to show on MESS like this. MESS missing something? Anyway I'm trying this out. I did notice the sounds aren't sounding the same as well. Maybe I have it on a different sounds sample settings.
Expansion 2 slots and later on the Hitachi HD68HC000 10 MHz model is 4 slots. Then back down to 2 slots afterward. I guest 6 slots having loading issues so that why they drop back to 4 slots.
Looking at the Sharp X68000 - Games (TOSEC-v2007-08-22_CM) games list. There are some that have anywhere 5 to 9 Disk.

I'm not familiar with the X68000 or X68K but it looks like that screen could be part of the emulator itself?
Either way MT-32, SC-55 etc are external MIDI synth modules and you'll need the ROMs for those as well I guess. This also explains why X68K and MESS sound different to you.

I found two clones that have the correct sounds.
WinX68k High-Speed and XM6 Pro-68k But these have issues that I found. Win X68K seem less buggy.

This is pretty common with floppy disk based (16 bit) computers. A plain Amiga 500 has 1 (internal) floppy drive and so much games came on 2/3 & more disks... ...Simply put first disk in drive and when You needed to, Amiga will "tell" You to insert disk 2/3 and so on...

That menu is to choose between normal X68000 internal sound (YM2151,MSM6258) and MIDI (MT-32 and SC-55 are the common ones). MIDI devices are not emulated yet, so the menu does not appear.

The X68000 comes with 2 floppy drives, and supports up to 4 (the limit of the FDC). Most games will only assume there are two though, but generally you can put the first two disks in the first two drives. Any disk labelled "System" is likely to be the bootable disk, so start with the system disk and data 1, and if it wants you to change a disk, it will tell you.
Definitely part of the game. Try booting Castlevania Chronicles on PS1. If you have the Japanese version, boot it into original mode. If you have the US version, boot it into original mode while holding L1 and R1.
That menu will appear and let you choose the sound system; the PS1 version includes digital recordings of all three sound hardware types.
